ABSTRACT

SUMMARY: We investigated the expression and clinical significance of miR-15b-5p in clear cell renal cell carcinoma (RCC) through bioinformatics analysis and experimental verification. The differentially expressed miRNAs were screened in the GEO database. Venn diagram showed that there were 5 up-regulated miRNAs (has-miR-210, has-miR-142-3p, has-miR-142-5p, has-miR-15b-5p, and has-miR-193a-3p) and only 1 down-regulated miRNA (has-miR-532-3p) that were commonly expressed between GSE189331 and GSE16441 datasets. This was further confirmed in TCGA. Further analysis showed that the has-miR-193a-3p, has-miR-142-3p, has- miR-142-5p, and has-miR-15b-5p were closely related to tumor invasion, distant metastasis and survival probability. The expression of miR-15b-5p in ccRCC tissues was significantly higher than that in adjacent normal kidney tissues (P0.05). Following inhibition of miR-15b-5p expression, RCC cells had attenuated proliferation, increased apoptosis, and attenuated migration and invasion. has-miR-15b-5p-WEE1, has-miR-15b-5p-EIF4E, has-miR-15b-5p-PPP2R1B may be three potential regulatory pathways in ccRCC. miR-15b-5p is highly expressed in cancer tissues of ccRCC patients. It may promote proliferation, inhibit apoptosis and enhance cell migration and invasion of RCC cells. The has-miR-15b-5p-WEE1, has-miR-15b-5p-EIF4E, and has-miR-15b-5p-PPP2R1B may be three potential regulatory pathways in ccRCC.


Investigamos la expresión y la importancia clínica de miR-15b-5p en el carcinoma de células renales (CCR) de células claras mediante análisis bioinformático y verificación experimental. Los miARN expresados diferencialmente se examinaron en la base de datos GEO. El diagrama de Venn mostró que había 5 miARN regulados positivamente (has-miR-210, has-miR-142-3p, has-miR-142-5p, has-miR-15b-5p y has-miR-193a-3p). ) y solo 1 miARN regulado negativamente (has-miR-532-3p) que se expresaron comúnmente entre los conjuntos de datos GSE189331 y GSE16441. Esto fue confirmado aún más en TCGA. Un análisis más detallado mostró que has-miR-193a-3p, has-miR-142-3p, has- miR-142-5p y has-miR-15b-5p estaban estrechamente relacionados con la invasión tumoral, la metástasis a distancia y la probabilidad de supervivencia. La expresión de miR-15b-5p en tejidos ccRCC fue significativamente mayor que la de los tejidos renales normales adyacentes (P 0,05). Puede promover la proliferación, inhibir la apoptosis y mejorar la migración celular y la invasión de células RCC. has-miR-15b-5p-WEE1, has- miR-15b-5p-EIF4E y has-miR-15b-5p-PPP2R1B pueden ser tres posibles vías reguladoras en ccRCC.

ABSTRACT

Objective To explore hemodynamics of the aortic arch and supraarch vessels after thoracic endovascular aortic repair with fenestration and parallel grafts techniques, and compare the differences of these techniques. Methods Four patients with aortic arch lesions whose supraarch vessels were reconstructed by different surgical techniques (fenestration, chimney and periscope) were studied, and three-dimensional (3D) geometric models were established based on postoperative image data. The physiological flow obtained from two dimensional (2D) phase contrast magnetic resonance imaging were imposed on the ascending aorta inlet and the supraarch vessels outlets. The pressure waveform of 3-element Windkessel model was imposed on the descending aorta outlet. Through computational fluid dynamics ( CFD ) simulations, the hemodynamic parameters were obtained, including the pressure of supraarch vessels, the velocity vector of the stent inlet, and the relative residence time. Results The pressure change of the periscope stent was the largest, followed by the fenestration stent, and the pressure change of the chimney stent was the smallest. The velocity of the fenestration and periscope stent inlet was uneven, which might form vortex. The velocity of the chimney stent inlet was even. The high relative residence time concentrated in distal end of the fenestration stent outer wall, the ‘gutter’ part, and the place where the chimney and periscope stent adhered to the vessel wall. Conclusions The pressure difference between the inner and outer walls of the fenestration and periscope stent was high, so it was recommended to use the balloon-expandable stent. The pressure difference between the inner and outer walls of the chimney stent was low, so it was recommended to use the self-expanding stent. The predicted location of thrombosis was consistent with the clinical follow-up data, so it may be used for surgical planning and risk assessment of interventional treatment of aortic arch lesions.

ABSTRACT Introduction Football is a sport with good acceptance among contemporary university students; it integrates competition, entertainment, and physical fitness. However, few studies demonstrate the concrete advantages found in college players. Objective Explore the impact of football on the physical performance of university students. Methods 100 university volunteers in elementary class 1-3 soccer practitioners, aged 19.2±2.1 years, were evaluated by a battery of tests in body composition, body function, and physical fitness. The results were compared with the literary data, and the experiment went through a mathematical, statistical method to separate the soccer intervention and conduct empirical research on physical health. Results After the 18-week elective course intervention, the 50m and 1000m sprint results changed significantly, especially the 1000m sprint performance greatly improved; the difference was significant (P<0.01). Conclusion Soccer is a sport that improves the physical function and quality of university students and is more conducive to maintaining the physical health of university students. RESUMO Introdução O futebol é um esporte com boa aceitação entre os universitários contemporâneos, integra a competição, o entretenimento e a aptidão física. Porém há poucos estudos demonstrando as vantagens concretas encontradas nos jogadores universitários. Objetivo Explorar o impacto do futebol no desempenho físico dos universitários. Métodos Um total de 100 universitários voluntários na classe fundamental 1-3 praticantes de futebol, com idade de 19.2±2.1 anos, foram avaliados por bateria de testes em áreas de composição corporal, função corporal e aptidão física. Os resultados foram confrontados com os dados literários, e o experimento passou por método estatístico matemático, para separar a intervenção do futebol e realizar pesquisas empíricas sobre a saúde física. Resultados Após a intervenção de curso eletivo de 18 semanas, os resultados de corrida de 50 metros e 1000 metros mudaram significativamente, especialmente o desempenho da corrida de 1000 metros melhorou muito, a diferença foi muito significativa (P<0,01). Conclusão O futebol é um esporte que melhora a função física e a qualidade dos universitários, além de validar-se como mais propício para a manutenção da saúde física dos universitários. RESUMEN Introducción El fútbol es un deporte con buena aceptación entre los estudiantes universitarios contemporáneos, que integra la competición, el entretenimiento y la forma física. Sin embargo, hay pocos estudios que demuestren las ventajas concretas encontradas en los jugadores universitarios. Objetivo Explorar el impacto del fútbol en el desempeño físico de los estudiantes universitarios. Métodos Un total de 100 voluntarios universitarios en el grado elemental 1-3 que practicaban el fútbol, con una edad de 19,2±2,1 años, fueron evaluados mediante una batería de pruebas en áreas de composición corporal, función corporal y aptitud física. Los resultados se confrontaron con los datos de la literatura, y el experimento pasó por el método estadístico matemático, para separar la intervención del fútbol y realizar una investigación empírica sobre la salud física. Resultados Después de la intervención del curso electivo de 18 semanas, los resultados de la carrera de 50 metros y de 1000 metros cambiaron significativamente, especialmente el rendimiento de la carrera de 1000 metros mejoró mucho, la diferencia fue muy significativa (P<0,01). Conclusión El fútbol es un deporte que mejora la función y la calidad física de los estudiantes universitarios, y se valida como más propicio para mantener la salud física de los estudiantes universitarios. ABSTRACT

Cervical cancer is a common malignant tumor that threatens women's life and health. Among them, the prognosis of recurrent or advanced cervical cancer is poor, and such patients often receive surgical treatment, radiotherapy, chemotherapy, etc., which makes it more difficult to choose a treatment plan. Even after the most effective treatment methods, the treatment failure rate of advanced cervical cancer is still high. Thanks to the research progress of tumor cell signaling pathway-related targets and immunotherapy, especially immune checkpoint inhibitors, new breakthroughs have been made in cervical cancer maintenance therapy. Targeted maintenance therapy, immune maintenance therapy, and multi-drug combination maintenance therapy are currently popular areas of cervical cancer maintenance therapy.

BACKGROUND: Acidithiobacillus ferrooxidans is a facultative anaerobe that depends on ferrous ion oxidation as well as reduced sulfur oxidation to obtain energy and is widely applied in metallurgy, environmental protection, and soil remediation. With the accumulation of experimental data, metabolic mechanisms, kinetic models, and several databases have been established. However, scattered data are not conducive to understanding A. ferrooxidans that necessitates updated information informed by systems biology. RESULTS: Here, we constructed a knowledgebase of iron metabolism of A. ferrooxidans (KIMAf) system by integrating public databases and reviewing the literature, including the database of bioleaching substrates (DBS), the database of bioleaching metallic ion-related proteins (MIRP), the A. ferrooxidans bioinformation database (Af-info), and the database for dynamics model of bioleaching (DDMB). The DBS and MIRP incorporate common bioleaching substrates and metal ion-related proteins. Af-info and DDMB integrate nucleotide, gene, protein, and kinetic model information. Statistical analysis was performed to elucidate the distribution of isolated A. ferrooxidans strains, evolutionary and metabolic advances, and the development of bioleaching models. CONCLUSIONS: This comprehensive system provides researchers with a platform of available iron metabolism-related resources of A. ferrooxidans and facilitates its application.

ABSTRACT Introduction: The formation and propagation of pulse waves are mainly accomplished by coordinating the heart and the vascular system. The contraction and relaxation of the heart are the sources of pulse waves. The aorta vibrates regularly as the heart contracts. This vibration propagates forward along the elastic blood vessel to form a pulse wave. The pulse wave contains very rich physiological and pathological information about the cardiovascular system. If there is a problem with the heart's structure, it can cause abnormal pulse waveforms. Objective: This article analyzes pulse waveform changes and blood flow during high-intensity interval training. It combines the test results to guide the exercise rehabilitation treatment of patients with chronic diseases. Methods: Pulse waves were collected from subjects under different exercise loads and the characteristics of pulse wave parameters under intermittent exercise were studied. Results: An athlete's pulse wave response is different in the case of high-intensity intermittent exercise. There are differences in the cardiovascular response of patients with different body weights. Conclusion: High-intensity interval training can improve the cardiovascular function of patients with chronic diseases and affect their pulse waveform. RESUMO Introdução: A formação e propagação de ondas dos batimentos cardíacos resultam principalmente da coordenação entre o coração e o sistema vascular. A contração e o relaxamento do coração produzem ondas de batimento cardíaco. A aorta vibra regularmente a medida em que o coração se contrai. A vibração se propaga no vaso sanguíneo elástico para formar uma onda de batimento cardíaco. Esta onda contém informações fisiológicas e patológicas muito ricas sobre o sistema cardiovascular. Se há algum problema com a estrutura cardíaca, ondas anormais podem resultar. Objetivo: Este estudo analisa as alterações em ondas de batimentos cardíacos e o fluxo sanguíneo durante treinos de intervalos de alta intensidade. Combina os resultados dos testes para orientar o tratamento de exercícios de reabilitação em pacientes com doenças crônicas. Métodos: Ondas de batimentos cardíacos foram coletadas de indivíduos praticando exercícios com cargas diferentes. As características dos parâmetros das ondas de batimento cardíaco relacionadas a exercícios intermitentes foram estudadas. Resultados: A resposta das ondas de batimentos de atletas é diferente em exercícios intermitentes de alta intensidade. Existem diferenças na resposta cardiovascular de pacientes com diferentes pesos corporais. Conclusão: O exercício de intervalos de alta intensidade pode melhorar a função cardiovascular de pacientes com doenças crônicas e afetar as ondas de batimentos cardíacos. La contracción y el relajamiento del corazón producen ondas de latido cardíaco. La aorta vibra regularmente a la medida que el corazón se contrae. La vibración se propaga en el vaso sanguíneo elástico para formar una onda de latido cardíaco. Esta onda contiene informaciones fisiológicas y patológicas muy ricas sobre el sistema cardiovascular. Si hay algún problema con la estructura cardíaca, puede causar ondas anormales. Objetivo: Este estudio analiza las alteraciones en ondas de latidos cardíacos y el flujo sanguíneo durante entrenamientos de intervalos de alta intensidad. Combina los resultados de las pruebas para orientar el tratamiento de ejercicios de rehabilitación en pacientes con enfermedades crónicas. Métodos: Ondas de latidos cardíacos se recogieron de individuos practicando ejercicios con carga diferentes. Las características de los parámetros de las ondas de latido cardíaco relacionadas a ejercicios intermitentes se estudiaron. Resultados: La respuesta de las ondas de latidos de atletas es diferente en ejercicios intermitentes de alta intensidad. Existen diferencias en la respuesta cardiovascular de pacientes con diferentes pesos corporales. Conclusión: El ejercicio de intervalos de alta intensidad puede mejorar la función cardiovascular de pacientes con enfermedades crónicas y afectar las ondas de latidos cardíacos. ABSTRACT

Objective Based on ergonomics and biomechanics simulation technology, the biological evaluations of human muscles and the rehabilitation strategies of foot rehabilitation robot with adjustable swing were carried out and studied. Methods The human-robot coupling model of human body and foot rehabilitation robot with adjustable swing were established by using AnyBody software. Through kinematics simulation of the coupling model, the comparison between the simulation result and the theoretical calculation result confirmed reliability of the coupling model. The parameter study of AnyBody software was used to perform the biomechanical simulation of the verified coupling model. By regarding velocity and swing of the foot rehabilitation robot as variables, the muscle activity and muscle force under different combinations of variables were analyzed. ResultsDuring rehabilitation exercise, stretching performance of the foot related-muscles was effectively trained. The influence of different velocity and swing amplitude on muscles was different, and safety range of the velocity and swing adjustment was obtained. Conclusions The combined analysis of muscle activity and muscle force under different velocity and swing was achieved. The results have certain guiding significance on clinical application of foot rehabilitation robot and formulation of rehabilitation strategies in passive rehabilitation mode.

@#[摘 要] 目的:探讨circ_0072088对甲状腺癌IHH4细胞恶性生物学行为的影响及其可能的机制。方法:选取2015年4月至2019年3月于青海大学附属医院就诊的确诊为甲状腺癌后接受切除手术且术前未进行任何治疗的48例甲状腺癌患者的甲状腺癌组织及其对应癌旁组织。根据GEO数据集(GSE93522)分析鉴定甲状腺乳头状癌中差异表达的circRNA。用qPCR法检测circ_0072088在甲状腺癌组织和细胞中的表达水平。采用CCK-8法和Transwell法检测circ_0072088和miR-532-3p过表达对甲状腺癌IHH4细胞增殖、迁移和侵袭的影响。采用生物信息学和双荧光素酶报告基因实验验证甲状腺癌细胞中circ_0072088与miR-532-3p、miR-532-3p和WEE1基因之间的关系。结果:circ_0072088在甲状腺癌组织和细胞中呈高表达(P<0.05或P<0.01)。circ_0072088过表达促进了甲状腺癌细胞的增殖、迁移和侵袭(P<0.05或P<0.01),而转染 miR-532-3p模拟物能够减弱这种作用(P<0.05或P<0.01)。此外,机制研究表明,circ_0072088可以与miR-532-3p靶向结合,且WEE1是miR-532-3p的下游靶基因。结论:circ_0072088通过调控miR-532-3p/WEE1轴促进IHH4细胞的增殖、迁移和侵袭。

@#[Abstract] Objective: To investigate the effects of antibiotics on the treatment efficacy of immune checkpoint inhibitors in NSCLC (non-small cell lung cancer) with Meta-analysis. Methods: Literatures regarding the effects of antibiotics on the treatment efficacy of immune checkpoint inhibitors in NSCLC were searched in Pubmed, Cochrane Library, Embase, EBSCO, Chinese Biomedical Literature Database(CBM) and Chinese Journal Full-text Database(CNKI). RevMan 5.3 software was used in this Meta-analysis. Results: Fourteen articles involving 2 505 NSCLC patients were included in this study. Meta-analysis showed that the application of antibiotics could significantly shorten the PFS (HR=1.14, 95%CI =1.04-1.26, P=0.005) and OS (HR=1.30, 95%CI =1.14-1.47, P<0.0001) of NSCLC patients treated with immune checkpoint inhibitors. Conclusion: Application of antibiotics before, concurrently or after immune checkpoint inhibitors in the treatment of NSCLC may significantly shorten PFS and OS, resulting in adverse effect on treatment efficacy.

ABSTRACT

Objective: To investigate the middle ear function of the patients with cleft palate pre and post palatoplasty. Methods: 76 patients with cleft palate were investigated by clinical history and audiology examinations including electric otoscopy,tympanometry and click-ABR threshold. Results: The risk for middle ear function decreased with advancing age in the first 5 years. It was noticed that the otologic outcomes was related to the CP type. During long time follow-up, the frequency with the middle ear function disorder was always high within the CP patients but the proportion of the patients received tympanostomy tubes was low relatively. The prevalence of middle ear dysfunction did not differ with the time of cleft palate repair. Conclusion: The patients with cleft palate have middle ear function dysfunction in a long period of time,therefore a standard long-time follow-up system is necessary.

ABSTRACT

Objective:To explore the protective effect of ulinastatin on sepsis-related acute lung injury (ALI) in mice and its related mechanism analysis.Methods:A total of 50 male mice aged 6-8 weeks were selected and randomly divided into 5 groups, with 10 mice in each group. Group A was the sham operation group, group B was the sham operation and ulinastatin (30 000 U/kg) intervention group, group C was the simple cecal ligation and perforation (CLP) group, group D was the CLP+low-dose ulinastatin (15 000 U/kg) treatment group, and group E was the CLP+high-dose ulinastatin (30 000 U/kg) treatment group. Ulinastatin in groups B, D and E was injected intraperitoneally at the corresponding dose 1 h before CLP operation. The hematological characteristics, lung edema and inflammatory changes of lung tissue in mice of each group were evaluated and the expression levels of interleukin(IL)-6, tumor necrosis factor(TNF)-ɑ, IL-1β and nuclear factor(NF)-κB p65 mRNA and protein expression in blood and lung tissue were compared among the five groups.Results:Compared with that in group A, the lung water content in group C was significantly increased: (78.68 ± 1.85)% vs. (51.98 ± 0.77)%, P<0.01. Compared with that in group C, pulmonary edema of the mice treated with ulinastatin was significantly reduced, with a certain dose-effect relationship. The wet weight/dry weight ratio of the lung tissue in group C was significantly higher than that of group A, B, D, E: 4.74 ± 0.28 vs. 2.23 ± 0.16, 2.25 ± 0.22, 2.89 ± 0.31, 2.09 ± 0.12, P<0.01, and group E had the lowest ratio. After ulinastatin intervention, the inflammatory manifestations of the lungs were reversed in a dose-dependent manner. The degree of structural destruction was improved, and edema and polymorphonuclear neutrophil infiltration were reduced. Ulinastatin could significantly reduce the neutrophil and lymphocyte counts of mice after CLP treatment, reduce the expression of IL-6, TNF-α, IL-1β inflammatory cytokines and MPO activity. The reverse transcription-polymerase chain reaction (RT-PCR) results and Western blot analysis showed that the mRNA and protein expressions of NF-κB p65 in group C of lung tissue was significantly increased. However, after treatment with ulinastatin, it was significantly reduced, and it had a certain dose-effect relationship. Conclusions:Ulinastatin pretreatment can significantly reduce the expression levels of IL-6, IL-1β and TNF-α in sepsis-related acute lung injury, and can alleviate the inflammatory response in the lung. The protective effect of ulinastatin may be related to the inhibition of NF-κB pathway activation.

ABSTRACT

Myeloid-derived suppressor cells (MDSCs) are newly discovered cells derived from bone marrow that suppress immunity, playing an immunosuppressive role in tumors, infections, and autoimmune diseases. It has been confirmed in disease model studies that their immunosuppressive effect is mainly achieved by suppressing the function of T cells. At present, the treatment of infectious diseases is still a major obstacle, especially in the treatment of chronic infectious diseases. Studies have found that MDSCs have increased aggregation in infectious diseases. As MDSCs are gaining more attention in infectious diseases, their potential therapeutic effects may be further explored. This article mainly reviews the immunosuppressive mechanisms of MDSCs in infections such as bacteria, viruses, parasites and fungi and their relationship with the diseases.

Psoriasis is an immune-mediated chronic inflammatory disease,whose occurrence is closely associated with genes,immunity,environment and other factors.Classic comorbidities associated with psoriasis mainly include cardiovascular diseases,obesity and inflammatory bowel diseases.Emerging data have shown some other diseases closely associated with psoriasis,including metabolic syndrome,chronic renal diseases,tumors,infections,and so on.This review summarizes psoriasis-associated comorbidities that have been reported in studies,and analyzes the possible co-pathogenesis of these diseases and psoriasis.

ABSTRACT

Objective:To investigate the correlation between serum uric acid level and non-alcoholic liver disease (NAFLD).Methods:A medical examination cohort of our hospital′s staff was constructed. From February 1, 2012 to January 29, 2013, a total of 3, 479 staff without NAFLD were selected as research subjects, and baseline data were collected through a questionnaire survey, physical examination, abdominal ultrasound examination, blood lipid, blood glucose, liver and kidney function, and other laboratory tests. From February 1, 2013 to December 31, 2018, the patients were followed up during the annual physical examination for six years. The serum uric acid level was used as the observation index and divided into four groups from A to D according to the quartile. With the occurrence of NAFLD as the outcome indicator; the four groups of uric acid as the observation indicator; and age, body mass index, hyperlipidemia, hyperglycemia, hypertension, creatinine, and alanine aminotrans ferase as confounding factors; four Cox regression analysis models were constructed to explore the relationship between groups of different blood uric acid levels and NAFLD. Stratified by gender, three Cox regression analysis models were constructed to investigate the relationship between blood uric acid level grouping and NAFLD between different genders.Results:The six-year cumulative incidence of NAFLD in groups A, B, C, and D was 1.2%, 3.1%, 4.9%, and 12%, respectively (χ 2=114.710, P<0.05). Among the female workers, the six-year cumulative incidence rates in groups A, B, C, and D were 1.0%, 2.9%, 4.1% and 10.9%, respectively (χ 2=71.241, P<0.05). The incidence risk of NAFLD in groups B, C, and D was 2.04 (1.01-4.11), 2.24 (1.13-4.44), and 3.89 (1.94-7.80) times that of group A, P<0.05, respectively. The incidence risk of NAFLD in groups B, C, and D was 2.21 (1.02-4.77), 2.39 (1.10-5.19), and 4.49 (1.99-10.15) times that of group A, all P<0.05, respectively. Conclusion:The risk of NAFLD increased with the increase of serum uric acid level, and this trend was mainly manifested in female employees.

ABSTRACT

BACKGROUND: Eugenol is an economically favorable substrate for the microbial biotransformation of aromatic compounds. Coniferyl aldehyde is one kind of aromatic compound that is widely used in condiment and medical industries; it is also an important raw material for producing other valuable products such as vanillin and protocatechuic acid. However, in most eugenol biotransformation processes, only a trace amount of coniferyl aldehyde is detected, thus making these processes economically unattractive. As a result, an investigation of new strains with the capability of producing more coniferyl aldehyde from eugenol is required. RESULTS: We screened a novel strain of Gibberella fujikuroi, labeled as ZH-34, which was capable of transforming eugenol to coniferyl aldehyde. The metabolic pathway was analyzed by high-performance liquid chromatography­mass spectrometry and transformation kinetics. The culture medium and biotransformation conditions were optimized. At a 6 h time interval of eugenol fed-batch strategy, 3.76 ± 0.22 g/L coniferyl aldehyde was obtained, with the corresponding yield of 57.3%. CONCLUSIONS: This work improves the yield of coniferyl aldehyde with a biotechnological approach. Moreover, the fed-batch strategy offers possibility for controlling the target product and accumulating different metabolites

ABSTRACT

BACKGROUND: This paper presents micro- and nano-fabrication techniques for leachable realgar using the extremophilic bacterium Acidithiobacillus ferrooxidans (A. ferrooxidans) DLC-5. RESULTS: Realgar nanoparticles of size ranging from 120 nm to 200 nm were successfully prepared using the highenergy ball mill instrument. A. ferrooxidans DLC-5 was then used to bioleach the particles. The arsenic concentration in the bioleaching system was found to be increased significantly when compared with that in the sterile control. Furthermore, in the comparison with the bioleaching of raw realgar, nanoparticles could achieve the same effect with only one fifth of the consumption. CONCLUSION: Emphasis was placed on improving the dissolvability of arsenic because of the great potential of leachable realgar drug delivery in both laboratory and industrial settings

ABSTRACT

Amyotrophic lateral sclerosis (ALS) is among the most common type of motor neuron diseases, and its pathogenesis remains unclear. In recent years, our understanding of the genetic basis of ALS has led to the development of various ALS disease models, which allow for screening of ALS-related drugs and treatment methods. This review focuses on the research progress of ALS, summarizes the systems of commonly used experimental animal models, including transgenic animals, gene knockout approaches and autonomous animal models, points to the problems needing attention in standardized ALS non-clinical research, and proposes the criteria for selection of standardized R&D model.

ABSTRACT

@# Objective: To explore the effect of miR-195/TLR4 axis on the proliferation, invasion and migration of liver cancer cells via regulating NF-κB pathway. Methods: Twenty-five pairs of liver cancer tissues and corresponding adjacent tissues surgically resected at the Second Affiliated Hospital of Kunming Medical University from March 2016 to January 2017 were collected for this study. Liver cancer HepG2 cells were cultured and then randomly divided into four groups: control group (NC), miR-195 mimic group (miR-195), TLR4 knockdown group (si-TLR4), and miR-195 inhibitor combined with TRL4 knockdown group (si-TLR4+miR-195 inhibitor). qRTPCR was used to detect the expression of miR-195 in liver cancer tissues and cell lines. CCK-8 assay was used to evaluate the cell viability of each group. Transwell and Wound healing assay were applied to detect the invasion and migration ability of HepG2 cells, respectively. Dual-luciferase reporter gene assay was used to verify the targeted regulation of TLR4 by miR-195. WB was applied to analyze the protein expressions of TLR4 and NF-κB p65. Results: miR-195 was down-regulated in the liver cancer tissues compared with adjacent tissues (P<0.01). Compared with human hepatic epithelial cells (THLE-3), the expression of miR-193 in liver cancer cell lines (HepG2 and Huh-7) was down-regulated (P<0.01), and the expression level in HepG2 cells was the lowest. The proliferation, invasion and migration of HepG2 cells was significantly suppressed after over-expression of miR-195 (all P<0.01). Moreover, over-expression of miR-195 significantly down-regulated TLR4 protein expression (P<0.05), and TLR4 was negatively correlated with miR-195 (R2= 0.602, P<0.0001). Furthermore, miR-195 over-expression inhibited proliferation, invasion and migration of HepG2 cells by targeting TLR4 expression and blocking NF-κB pathway (P<0.05 or P<0.01). Conclusion: miR-195 over-expression can inhibit the proliferation, invasion and migration of HepG2 cells. The mechanism may be related with targeting TLR4 and blocking the NF-κB pathway to affect cell biological behaviors.

ABSTRACT

Psoriasis is a chronic,non-infective inflammatory dermatosis characterized by erythemas covered with scales.Psoriasis is considered as a systemic disease,and several studies have confirmed that it is associated with many other diseases,such as cardiovascular diseases,obesity,diabetes mellitus type Ⅱ,dyslipidemia,nonalcoholic fatty liver disease,anxiety disorder,depression and inflammatory bowel disease.The occurrence and development of psoriasis is closely related to the immune response,and the immune dysfunction of T lymphocytes induced by the infiltration and activation of inflammatory cells has been confirmed as the key step in the pathogenesis of psoriasis.Various kinds of cytokines in the local microenvironment can promote the activation and proliferation of T lymphocytes,and regulate the differentiation of T lymphocytes into different subsets,which take an important part in the occurrence of psoriasis.In recent years,researchers have found that local microbes also play an important role in this process.Thus,this review summarizes the progress in the roles of human skin and gut microbiota in psoriasis.

ABSTRACT

BACKGROUND: Histone acetylation is an important epigenetic modification that regulates gene activity in response to stress. Histone acetylation levels are reversibly regulated by histone acetyltransferases (HATs) and histone deacetylases (HDACs). The imperative roles of HDACs in gene transcription, transcriptional regulation, growth and responses to stressful environment have been widely investigated in Arabidopsis. However, data regarding HDACs in kenaf crop has not been disclosed yet. RESULTS: In this study, six HDACs genes (HcHDA2, HcHDA6, HcHDA8, HcHDA9, HcHDA19, and HcSRT2) were isolated and characterized. Phylogenetic tree revealed that these HcHDACs shared high degree of sequence homology with those of Gossypium arboreum. Subcellular localization analysis showed that GFP-tagged HcHDA2 and HcHDA8 were predominantly localized in the nucleus, HcHDA6 and HcHDA19 in nucleus and cytosol. The HcHDA9 was found in both nucleus and plasma membranes. Real-time quantitative PCR showed that the six HcHDACs genes were expressed with distinct expression patterns across plant tissues. Furthermore, we determined differential accumulation of HcHDACs transcripts under salt and drought treatments, indicating that these enzymes may participate in the biological process under stress in kenaf. Finally, we showed that the levels of histone H3 and H4 acetylation were modulated by salt and drought stress in kenaf. CONCLUSIONS: We have isolated and characterized six HDACs genes from kenaf. These data showed that HDACs are imperative players for growth and development as well abiotic stress responses in kenaf.
